Dietary supplementation with lipoic acid for the prevention of obesity and insulin-resistance: study of the mechanisms involved. pedro luis prieto-hontoria faculty of pharmacy. University of navarra (spain). 2011 the aim of this study was to investigate the effects of dietary supplementation with lipoic acid (la) on body weight gain, adiposity, and insulin sensitivity in control and high fat fed rats. The changes induced by la on several adipokines involved in the regulation of body weight and glucose homeostasis were also tested both in vivo and in different models of cultured adipocytes. la supplementation induced a lower body weight gain and adipose tissue size in rats accompanied by a reduction in food intake. Moreover, la reduced feed efficiency and decreased intestinal α-methylglucoside absorption. Dietary supplementation with la also reversed the insulin-resistance induced by the high fat diet and upregulated adiponectin gene expression and ampk phosphorylation in white adipose tissue. In 3t3-l1 adipocytes, la also activated ampk, but caused a dose-dependent inhibition on adiponectin production. In addition, la caused a dose-dependent inhibition of leptin secretion which was related to increased anaerobic metabolism of glucose in 3t3-l1 adipocytes. The phosphorylation of sp1 and the reduced activity of this transcription factor are also likely to be involved in the inhibition of leptin induced by la in adipocytes. La also inhibited chemerin secretion and mrna levels in both 3t3-l1 adipocytes and in subcutaneous and omental adipocytes from obese subjects. Moreover, la was able to abolish the stimulatory effects of the pro-inflammatory cytokine tnf-α on chemerin secretion. In summary, these findings suggest that la could be a potential therapeutic agent for the treatment of some of the metabolic complications associated to obesity. keywords: lipoic acid, obesity, adipose tissue, adipokines
