Cell competition is a process through which unfit cells (losers) are replaced from a tissue by neighboring optimal cells (winners). Drosophila flower protein forms a code that serves to distinguish winners from losers and, thus, mediates the initial steps of cell competition. Flower is conserved from fungi to humans. Mouse flower encodes four protein isoforms. These mfwe proteins are membrane proteins whose exact function in the mouse is still unknown. To study mfwe function in mammals and its role during carcinogenesis, we generated a conditional and constitutive fwe knock-out mouse model. Fwe-deficient mice display a normal phenotype. However, deficiency of mfwe reduces the number of papillomas formed as a result of dmba/tpa carcinogenesis protocol, but does not influence tumorigenesis in other contexts of cancer such 3-mca and aom/dss chemical carcinogenesis or emu-myc, apcmin/+ and p53-/- genetic mouse models of cancer.
