This thesis examines a method to study the effect of long-acting neuroleptics on fear in sheep as a model for wild ungulates using the open-field test (oft) and the stress-induced hyperthermia (shi) caused by the oft. the consistency of individual behaviour in the oft and of sih caused by the oft was studied using data from two breeds of sheep tested repeatedly over time. Twenty four ewes of two breeds, lacaune and ripollesa, were tested in an arena measuring 5 m x 2.5 m. All animals were tested for 10 min and all behaviours were recorded throughout the test. Rectal temperature was measured immediately before the start of the test (t1) and 10 minutes after finishing it (t2). Sih was measured as the difference between t2 and t1. Sheep were tested over three periods including three experimental days each. Ewes showed consistent changes in behaviour in the oft and a clear sih response. Breed differences were found in sih, lacaune ewes having a lower sih than ripollesas. The same animals were tested on days 1, 2, 3, 4, 7 and 9 after ppz injection. Ppz decreased bleats on days 2, 3, 4 and the sih response on days 2 and 3. Treated animals showed positive correlations between sih and bleats, squares entered and attempts to escape, as well as negative correlation between sih and visits to the food bucket. The effects of ppz validate the oft and sih as indicators of fear, as it reduces the number of bleats and the sih response, which are indicators of fear in sheep. a high performance liquid chromatographic (hplc) method for the determination of perphenazine (ppz) in sheep plasma was developed and validated. Using this method, the pharmacokinetic profile of ppz in sheep was studied and used as a model for other ungulates. The pharmacokinetic profile of ppz in dorcas gazelle (gazella dorcas) and investigated the effects of fluphenazine decanoate (fd) on normal behaviour in captive barbary sheep (ammotragus lervia) were evaluated. Perphenazine peak plasma concentration in dorcas gazelle was obtained 4 hours after administration, which is a time-lag similar to that found in sheep. Therefore, sheep may be a good model to study perphenazine enanthate in some wild ungulates. In barbary sheep, fd increased time spent active and reduced time spent feeding. The effects on activity were apparent the first week after fd administration, whereas effects on feeding appeared the second week after fd administration. Three weeks after fd administration behaviour returned to pre-administration values. The effects of fd in captive barbary sheep were different from those found in previous studies in other ungulates and further research is are needed to determine the factors explaining such variability.
